A detailed analysis of the genetic diversity of Xanthomonas oryzae pv. oryzae from disease epidemic areas in Tamil Nadu, Kerala, Karnataka and Andhra Pradesh was conducted. From a total of 132 infected leaf samples collected from 4 states, 96 bacterial strains were isolated. Identification of X. oryzae pv. oryzae was performed using gene-specific primer through which 50 strains of X. oryzae pv. oryzae were confirmed. Further, these strains were subjected to pathogenicity test on IR24 rice variety in which all 50 strains showed susceptibility to highly susceptible reactions. Genetic variation among the isolated X. oryzae pv. oryzae strains was studied using IS1112 DNA fingerprinting analysis. A dendrogram was derived by the UPGMA method which formed two clusters A and B. The cluster A had two subclusters I & II and cluster B had five subclusters I, II, III, IV and V.
