Combination of meristem tip culture and thermotherapy at 37 ± 2°C was very effectiv in eradicating potato virus X (PVX) in potato tubers depending upon the genotype. Despite some rottage/elongation of sprouts at 37 ± 2°C, maximum number of PVX-free plantlets were obtained in cv. Kufri Jyoti (95%) followed by Kufri Chandramukhi (85.4%) and Kufri Sindhuri (55.0%) respectively. Similarly, in PVX infected potato callus the concentration of PVX was reduced in all the cultivars tried. The reduction of PVX was maximum in cv. Kufri Jyoti held at constant temperature of 30 and 35°C whereas the PVX concentration was higher in all calli grown at alternate temperature.
