Buffer variants and detection of potato virus Y by penicillinase DAS-ELISA

Abstract

For detection of PVY in penicillinase (PNC) DAS-ELISA, an alternative buffer Ammonium bicarbonate (0.05M NH4HCO3, pH 8.5) was found compatible in all seven combinations with standard buffers (sodium carbonate, pH 9.8 & phosphate, pH 7.2) as coating, extraction and conjugate buffer. The sensitivity of test was highly stable in unwashed (tipped off) IgG coated plates upto overnight incubation period. The specificity was consistent in plates coated with IgG in alternate buffer and remained active up to 102 days in plates coated and stored at room temperature (14–20°C) after tipping off after 3 hrs incubation at 37°C.
