Isolation, Characterization and Sequencing of Lactobacillus from the Oral and Fecal Samples of Healthy Dogs

Abstract

Canine Oral and Fecal Lactic Acid Bacteria Strains were isolated from 40 healthy dog oral and fecal samples. Lactic Acid Bacteria were found in 70% of canine fecal samples and 40% of canine oral samples. Then these samples were cultured on Lactobacillus Selective Media with acetic acid. Out of these 6 samples were identified with partial 16s rRNA gene sequencing and various Biochemical Tests for their pro-biotic characteristic. Lactobacillus fermentum, Lactobacillus plantarum, Lactobacillus casei, Lactobacillus mucosae, Lactobacillus delbrueckii and Lactobacillus salivarius were selected as candidate pro-biotic strains based on the frequency and quantity in feces, growth density, acid tolerance, and antimicrobial activity. Sequence similarity values of 93% with Lactobacillus casei, 89% with Lactobacillus pentosus, and 85% with Lactobacillus delbruecki. These were the most frequently encountered bacteria in canine oral cavity and feces.
