Glipizide is a sulfonylurea used for the treatment of diabetes mellitus. Glipizide belongs to BCS class II which possess low solubility and high permeability which is a great disadvantage in administration of drug. One of the finest approaches to increase the solubility is preparation of nanoparticles. A simple formulation approach for preparation of nanosuspension of Glipizide is performed using HPMC K 15 M, PVP K 30, Eudragit L 100 as polymers and Poloxamer 188 as stabilizer by nano precipitation method. The formed nanosuspension was evaluated for various evaluation parameters like particle size, zetapotential, drug content and dissolution testing. The compatibility between the drug and ingredients are checked by using DSC. The best formulation containing the 2.0% of Eudragit L 100 showed the particle size of 98-107nm, Zeta potential of -55.3±1. The invivo studies of G12 formulation is conducted on Wistar rats for estimation of various pharmacokinetic parameters showed better profile when compared to pure drug. Hence nanoprecipitation was successfully employed for the preparation of Glipizide nanosuspension to improve the bioavailability.
