Abstract

To Develop and Validate Analytical method which is simple, rapid and sensitive for the determination of biotin from premixes (solid blend of multi-vitamin) by RP-HPLC. RP-HPLC analysis was performed using HPLC Agilent Technology 1260 Infinity series HPLC. The chromatographic separation was obtained by using column C18 Thermo scientific ODS Hypersil, 150×4.6, 3 μ, mobile phase used was buffer: acetonitrile in the ratio 91.5: 8.5, Flow rate 1.2 mi/min, wavelength 200nm, Injection volume 100 micro lit, column oven temp. 40°C, run time: 15 min. The retention time of biotin was found to between 10–12 min. Calibration curve was linear with a correlation coefficient of 0.996 over the concentration range of 50–150 ppm and precise with % RSD < 2 for standard and for sample % RSD < 5. The method was found to be specific, linear, precise and accurate. Suitable for the Quality control analysis.
