The aim of the study was to evaluate the post coital anti-implantation and antifertility studies of transdermal drug delivery of ethinylestradiol and medroxy progesterone acetate in wistar rats developed earlier in the laboratory. Medroxyprogesterone acetate (MPA), in combination with ethinylestradiol in a transdermal patch was developed and the patch was further evaluated for post coital anti implantation potential and antifertility studies.
The transdermal patch was prepared by solvent casting technique using various hydrophilic and hydrophobic polymers. The formulated patch was assayed using UV spectrophotometer using Vierordt's equation for ethinylestradiol and medroxy progesterone acetate. Anti-implantation and fertility studies were performed in female wistar rats. Anti-implantation and antifertility studies were performed in female wistar rats by counting the number of implantations and antifertility studies.
Pharmacodynamic studies of the formulated and developed transdermal patch in female wistar rats reflected 100% anti-implantation activity. Moreover, the overall amount of drug incorporated in the transdermal patch was less as compared to oral drug delivery since the first pass metabolism was bypassed in the transdermal patch. The developed optimized patch had excellent anti implanatation potential and showed evidence of good antifertility potential. These results provided evidence that the prepared patch should be further subjected to pharmacokinetic study.
The study concluded that there are high anti implantation and antifertility potential of the developed formulated patch containing ethinylestradiol and medroxyprogesterone acetate
