Simultaneous estimation of Rutin and Quercetin in Bidens pilosa, Convolvulus arvensis and Neurada procumbens by RP-HPLC

Abstract

The flavonoid compounds, namely rutin and quercetin were estimated simultaneously from Bidens pilosa, Convolvulus arvensis and Neurada procumbens and by reverse phase high-performance liquid chromatography (RP-HPLC). The compounds were eluted using methanol-water as a mobile phase by gradient program and detected at 254nm by a UV-Visible detector. The developed method showed well-resolved peaks of rutin and quercetin with the retention time of 15.032 minutes and 16.026 minutes respectively. The quantitative analysis showed that Bidens pilosa contains 4.73mg/g of rutin and 0.32mg/g of quercetin in the methanol extract. Convolvulus arvensis contains 18.30 mg/g of rutin and 0.21mg/g of quercetin whereas, Neurada procumbens contains 6.67mg/g of rutin and 4.51mg/g of quercetin in the methanol extract. Calibration curves were linear in relatively wide ranges of concentrations (0.1–600μg/ml for rutin and 0.1–300μg/mlfor quercetin) with high correlation coefficient values, r2 = 0.9970 and 0.9981 for rutin and quercetin respectively. The LOQ of rutin and quercetin were found to be 100ng/ml and LOD was found to be 35ng/ml. The method ascribed is simple, precise, sensitive and accurate for the simultaneous estimation rutin and quercetin in the plant extracts. This method could be conveniently and accurately used for the standardization or quality control of extracts of these plants.
