Fabrication and cytotoxic activity of sodium alginate ester nanoparticles loaded with clofarabine on MCF-7 cell linings

Abstract

Clofarabine is a second-generation purine nucleoside analogue with antineoplastic activity. The objective of this study was to prepare the Clofarabine nanoparticles loaded as OLEO-ALGINATE-ESTER conjugate, that OLEO-ALGINATE-ESTER conjugate could self-assemble to form nanoparticles when dissolved in water and the sparingly soluble Clofarabine could be entrapped within nanoparticles that stabilizes the Clofarabine in aqueous medium. Clofarabine loaded OLEO-ALGINATE-ESTER nanoparticles (Clofarabine-OLEO-ALGINATE-ESTER Nps) were prepared by simple ultra-sonication method. Meanwhile, the physiochemical characteristics of Clofarabine-OEA-Nps including particle-size and its distribution, zeta potential, drug entrapment efficiency and in-vitro drug release studies were determined. Furthermore, the cytotoxicity and cellular uptake of conjugated nanoparticles in to cancer cell linings were investigated.
