Analytical Method Development and Validation for the Quantification of Morin Hydrate in Nanocarriers by UV-Spectrophotometric and HPLC

Abstract

Morin hydrate, a bioflavonoid in the flavanol class, exhibits potent anti-inflammatory, antiarthritic, and anticancer properties. The present study focuses on the analytical quantification of morin hydrate in nanocarriers (MHNCs) by UV-Visible Spectrophotometry and High-Performance Liquid Chromatography (HPLC). UV-visible spectroscopy was used to generate a calibration curve for morin hydrate (MH) and Morin hydrate loaded nanocarriers (MHNCs), determining precision, accuracy, Limits of Detection (LOD), and Limits of Quantification (LOQ). Preformulation studies, including solubility, Differential scanning calorimetry (DSC), X-ray diffraction study (XRD), and Fourier-Transform Infrared spectroscopy (FTIR), confirmed the purity and physicochemical properties of morin hydrate. Morin hydrate and MHNCs were evaluated using UV-visible spectroscopy, revealing λ max at 265nm, concentration range of 5-25μg/ml, with the regression coefficient (R2) being 0.9971 and 0.997, respectively. HPLC was used to detect distinct peaks of morin hydrate and MHNCs under ambient conditions. The mobile phase consisted of methanol:0.1% orthophosphoric acid (pH 2.7) 75:25 as the mobile phase with a flow rate of 0.8ml/min. The temperature was kept at 25°C, and detection at 265nm was performed using a photodiode array (PDA) detector. The Resolution Time (Rt) for MH and MHNCs were 3.606±0.005 and 4.204±0.004, respectively. The theoretical plate (TP) values of 2354±96 and 3492±89, as well as the tailing factor (TF) values of 1.36±0.002 and 1.20±0.003, were within the specified constraints. The developed UV and HPLC method provide simple, precise, and consistent quantification of morin hydrate in a nanocarrier, which is crucial in enhancing drug delivery strategies and effectiveness in metabolic diseases like cancer, arthritis, diabetes treatment.
