Natural materials can be used as new sources of medicinal substances because of their content of secondary metabolites. In Indonesia, there are approximately 30000 species of plants containing secondary metabolites, with around 7500 species recognized as medicinal plants, one of which is ganitri (Eleocarpus ganitrus Roxb.). The aim of this study was to determine the total flavonoid content and antioxidant activity of the ethanol-water extract of ganitri leaves. The total flavonoid content was determined using UV-Vis spectrophotometry with AlCl3 complex reagent, while the antioxidant activity was assessed using the DPPH (2,2-diphenyl-1-picrylhydrazyl) and ABTS (2,2-azinobis-3-ethylbenzothiazoline-6-sulfonic acid) assays. The results showed that the total flavonoid content of the ethanol-water extract of ganitri leaves was 35.054±0.19mgQE/g. The antioxidant activity test results revealed that the extract had an IC50 of 5.04ppm according to the DPPH assay and 1.175ppm according to the ABTS assay, both of which indicate a very strong antioxidant activity.
