A selective and sensitive reverse phase high performance liquid chromatography (RP-HPLC) method has been developed for the separation and quantification of rabeprazole sodium and domperidone maleate in tablet dosages form has been fist developed and validated. Quantification carried out using HIQ Sil C18 column (25cm x 4.6mm i.d.) as a stationary phase and mobile phase comprised of 50mM KH2PO4 and acetonitrile in proportion of 60:40 (v/v) with pH adjusted to 7 ±1 by using 5M potassium hydroxide. The flow rate was 1.0 ml/min and monitored at 285 nm. The retention time for rabeprazole and domperidone were 6.707 and 8.002 min, respectively. The method was validated in terms of linearity, precision, accuracy, ruggedness, and specificity, limit of detection and limit of quantification. The linearity (r2) and percentage recoveries of rabeprazole and domperidone were 0.9986 and 100.02μg/ml and 0.9992 and 99.98% respectively. The proposed method is suitable for simultaneous determination of rabeprazole and domperidone in tablet dosages form.
