Formulation and Evaluation of Cefpodoxime Proxetil Matrix Tablets by Using Different Polymers

Abstract

Cefpodoxime proxetil is a third generation cephalosporin derivative used in various infectious diseases. It is practically insoluble in water. It is an orally administered drug with biological half life of 2–3 hours. Till date there is no controlled release tablets of this drug is available in the market. Thus controlled release matrix tablets was prepared and evaluated to provide controlled release of drug. The effects of some selected polymers on release profile of the drug from the systems have been investigated. The drug was fabricated with polymers such as HPMC (of various grades), ethyl cellulose, and guar gum in various ratios. Tablets were prepared by direct compression method and both pre-compressive and post-compressive parameters for all the batches, Short term accelerated stability studies, FT-IR Spectrophotometic analysis Dissolution studies were evaluated The dissolution data so obtained was fitted to various mathematical kinetic models and the drug release followed zero order and Higuchi's model. To study release mechanism of drug from matrices the data were fitted to Koresmeyer-peppas model and the release mechanism involved was non-Ficknian diffusion (Anomalous transportation). In-vitro release profile of cefpodoxime proxetil from various polymers showed a reduction in the release rate of drug (CFP 1 to CFP 9).by using higher viscous grades of polymers from HPMC K 4M to K 100 M Formulations containing Guar Gum in lower proportions gave better results compare to formulations containing HPMC or Ethyl cellulose as matrix materials..The data so obtained after the study reveals that Guar gum can be used in appropriate proportions to formulate matrix tablets of Cefpodoxime proxetil for controlled release of the drug for a period of 24 hours.
