Abstract

For improving bioavailability in controlled release system and to avoid the hepatic first pass metabolism of terbutaline sulphate, to develop a formulation of terbutaline sulphate in the form of mucoadhesive buccal film, which would provide prolong effect in relief of asthma. To achieve the goal of providing prolonged effect, buccal film were prepared by solvent casting technique using various mucoadhesive polymers like HPMC K4M, NaCMC alone and in combination with Carbopol 934P. All buccal film formulation were evaluated for their weight variation, thickness, folding endurance, surface pH, drug content, swelling index, water vapor transmission study, In-vitro bioadhesion strength, In-vitro residence time, In-vitro drug release study and Ex vitro permeation study. The film exhibited controlled release over more than 6hr. From the above study, it was concluded that buccal film containing 5mg Terbutaline sulphate in 3%w/v HPMC and 3%w/v sodium CMC exhibited moderate swelling, maximum In-vitro residence time, bioadhesion force as well as slower rate of In-vitro drug release which can be good way to bypass the extensive hepatic first pass metabolism and improve the bioavailability of Terbutaline sulphate.
