The present work was done to study the biological characteristic of colon cancer cells resistance to plant drug therapy. The extract of citrus reticulata essential oil was screened for their possible antioxidant activity by DPPH free radical scavenging and cytotoxicity study of proliferation on HT-29 colon cancer cell line was determined by using 3[4,5-dimethylthiazol-2-yl]-2,5-diphenyl tetrazolium bromide (MTT) microculture tetrazolium viability assay. The cells were exposed to different concentrations (100, 50, 25, 12.5, 6.25and 3.125 μg/ml). In DPPH radical scavenging assay the % inhibition at 100 μg/ml was 78.62 and % cytotoxicity in MTT assay at 100 μg/ml was 66.77 with IC50 of 17.56 and Rvalue of the extract 0.9962. From the above results it was observed that citrus reticulata essential oil extract of fruit peel part was more significant and could be consider as a potent sources of anticancer compounds for colon cancer activity.
