Abstract

Coprocessed directly compressible excipients by granulation technique in different ratios were prepared using plastic and brittle materials. Here coprocessing of microcrystalline cellulose and mannitol, microcrystalline cellulose and lactose, maize starch and mannitol, maize starch and lactose in the ratios 1:1, 1:3, 1:5 were prepared. The developed coprocessed excipients were studied for their influence on flow, strength of the tablet and dissolution characteristics of Donepezil HCl from direct compressible tablets. As the amount of brittle material increased the flow was found to be good. Optimized coprocessed formulation containing microcrystalline cellulose and mannitol in the ratio 1:5 was found to be more acceptable to formulate Donepezil HCl tablets. The preformulation parameters like flow property and the performance parameters were dependent on the nature and ratio of the diluents used for developing the coprocessed excipient.
