Production of L-Asparaginase by Marine Actinobacteria Isolated from the Rhizosphere of Mangrove Plants

Abstract

The aim of the present study was to isolate L-asparaginase producing actinobacteria from mangrove sediment. The samples were collected from Parangapattai coastal area (Lat.11°29′N Long.79°46′E) of Tamilnadu, India. A total of 24 actinobacteria colonies were isolated, among them 14 strains were separated and selected based on colony morphology. The isolated actinobacterial strains were screened for L-Asparginase enzyme production on M9 medium. Out of 14 isolates, 4 isolates (VITPPH1, VITPPH6, VITPPH7 and VITPPH10) showed positive results in Rapid plate Assay method. All the four actinobacterial strains showed 25–60 μg ammonia/mL/h range of L-asparagines activity. VITPPH10 strain showed highest L-asparaginase activity (58 μg ammonia/mL/h). Among the 14 isolates, VITPPH10 isolate showed best antimicrobial activity against Salmonella typhi and Pseudomonas aeruginosa. Cytotoxic assay was determined by using brine shrimp assay. VITPPH10 showed 100% mortality within 24 hours. The potential actinobacterial strain VITPPH10 was identified as Streptomyces sp.
