A simple, precise, rapid and reproducible reversed phase high performance liquid chromatography method is developed for the simultaneous estimation of glimepiride and metformin present in multicomponent dosage forms. Chromatography is carried out binary system on phenomenex C18 column (250 × 4.6 mm, 5 μm) with a mobile phase consisted of acetonitrile: acetate buffer (pH 4) in the ratio of 65:35, v/v at a flow rate of 1.0 ml/min. Detection is carried out using UV-PDA detector at 245 nm. The Rt for glimepiride and metformin are 5.709 ± 0.02 and 2.517 ± 0.05 minute respectively. The LOQ of glimepiride and metformin 0.038 and 0.221 μg/ml respectively. The RSD for three replicate measurements in three concentrations of samples in tablet are always less than 2%.
