Somatic embryogenesis and plant regeneration from shoot tip explants of Buchanania lanzan

Abstract

Plant regeneration through somatic embryogenesis was achieved from callus cultures derived from shoot tip explants of Buchanania lanzan. Somatic embryos formed on the surface of the embryogenic callus and, in some cases, directly from shoot tip explants. Callus cultures were initiated from shoot tip pieces of Buchanania lanzan on Murashige and Skoog medium supplemented with (1.0–2.0 mg/l) 2,4-D (2,4-dichlorophenoxyacetic acid), 0.50 mg/l Kinetin and 30 g/l sucrose. Maximum percentage response for callus formation was 62.22% on MS medium supplemented with 2,4-D (2.0 mg/l), Kn (0.50 mg/l) with PVP (100 mg/l). Somatic embryogenesis was achieved after transfer of embryogenic callus NAA (1.0 mg/l) with BAP (2.5 mg/l). The frequency of germination was 6.66% obtained on MS basal medium containing BAP (2.5 mg/l) combination of NAA (0.5 mg/l) 100 mg/l ascorbic acid with 30 g/l sucrose. Very low shoot initiation of 6.66% was observed in MS supplemented by BAP (5.0 mg/l) with NAA (0.5 mg/l) in combination with L proline (100 mg/l). The cut end of the micro-shoots were induced to root in vitro. The roots were obtained from MS + NAA 2.5 mg/l media.
