Abstract

Determination of genetic purity of seed lots is of significant importance to ensure a proper seed certification. A set of 20 DNA markers was used to generate a reliable marker haplotype that would serve as a reference guide to establish an identity of rice varieties bred from SKUAST-K. A marker haplotype that comprised of SSR markers namely, RM245, RM72, RM27933 and RM515 is proposed for characterization of indica rice varieties recommended for cultivation across temperate regions of Kashmir. The PCR assay made it possible to exercise hierarchical cluster analysis and helped to classify the varieties into japonica, indica and Basmati rice groups. DNA marker based fingerprint generated here may reliably be used for discrimination of varieties and also shall help to maintain varietal purity standards in commercial seed chain.
