Evaluation of effect of extenders on milt quality of Schizothorax niger (Heckel, 1838)
Abstract
This study aimed to assess the quality of milt of Schizothorax niger and analyse the effect of different extenders on it. Milt collected during spawning season from twenty male brooders having total length of 100–310 mm and body weight of 66–400 g were injected with calculated amount of hormone preparation Ovatide containing a synthetic analogue of Gonadotropin Releasing Hormone (GnRH) with dopamine antagonist @ 0.3 ml/kg of their body weight. Milt quality was determined by various sperm quality parameters such as milt volume, colour, consistency, pH, sperm motility and sperm concentration (spermatozoa/ml). Milt was preserved in ice cooled collection tubes at 4oC in refrigerator after diluting with three extenders and chemicals like 0.5% NaCl and 0.1% NaHCO3. For short-term preservation, milt was diluted in the ratio of 1:4 with different extenders/chemicals and maintained for 72 hrs in refrigerator. One of the extenders did not show good results.
