Shoot Organogenesis and Plantlet Regeneration from In Vitro Raised Leaf Segments of Jatropha curcas L.

Abstract

A simple protocol was developed for plantlet regeneration through organogenesis from leaf segments of Jatropha curcasL. Ten-day-old seedlings aseptically germinated on sand wet with water were used leaf and hypocotyl explant source. Initial incubation for 5 days in dark at 26 ± 2°C followed by transfer to 16/8-h light/dark cycle which lead to high germination of seeds and favored regeneration. The decisive role of 6-Benzyleaminopurine (BAP) at different concentrations was established on shoot organogenesis. Murashige and Skoog's (MS) medium fortified with 2.0 mg L−1 of BAP was optimum for shoot regeneration and addition of 1.5 mg L−1 IBA (Indole-3butric acid) was optimal for shoot elongation. The elongated shoots were rooted with 42.6% efficiency on half strength MS medium enriched with 1.0 mg L−1of IAA (Indole-3-acetic acid) which was further hardened in autoclave soil and transferred to green house chamber.
