Seven dried fish species were collected from local fish markets of Hyderabad and Mumbai, India to identify the species in mixed sample. PCR-RFLP technique was adopted using Bioanalyser to differentiate the selected fish species RFLP was carried out with three restriction enzymes viz., Dde1, HaeIII, NlaIII. The restriction fragment length pattern of each species was obtained. It was found that the 464 bp fragments of Cytochrome b gene in selected species showed divergence in the range of 5–10% among species. Bioanalyser 2100 facilitated rapid identification of dried fish species. RFLP decoder analysis resulted in efficient matching of species to the tested dried species. The study revealed that Bio analyser 2100 and its decoder technology could be effectively used at commercial level to perfectly identify dried fish which would avoid unscrupulous matching of species in local and international markets.
