Analysis of Genetic Diversity in Sesamum genotypes using Randomly Amplified Polymorphic DNA Markers

Abstract

Fourteen accessions of sesame (Sesamum indicum L.), an important oil seed crop of the tropics and subtropics were analysed using random amplified polymorphic DNA (RAPD) technique. Twenty six primers were tested in this study, out of which 20 revealed clear and repeatable RAPD polymorphisms. They produced 201 total loci and the total bands were 1780. Out of the 201 loci produced, 120 were polymorphic and hence the total polymorphism percentage was found tobe 59.70%. The average PIC value for RAPD was 0.86. The highest PIC value obtained was 0.91 for OPA-10 and OPK-13 markers and lowest PIC value was 0.63 for 17-ES. The number of amplicons produced per primer varied from 9 to 26 with a mean of 16.56 bands per primer. The molecular size of the amplified PCR products ranged from 230 to 6370 bp. On the basis of dendrogram, fourteen accessions were categorized mostly according to their colors.
