Optimization of Shooting and Rooting Media for in vitro Propagation on Standard Carnation (Dianthus caryophyllus L.) Cuttings

Abstract

The present experiment work involves shoot formation, their shoot multiplication and rooting in commercial standard varieties were Domingo (Red), Baltico (White) and Bizet (Pink) Carnation (Dianthus caryophyllus) during 2013–2014. For shoot formation both shoot tip and nodal cuttings were used. MS medium containing BAP alone or in combination with kinetin was tested. Best shoot formation response was obtained in Domingo after 7 to 8 days of inoculation from shoot tip on MS medium supplemented with (4.0 mg/l BAP & Kin) in nodal explant 9 to 11 days for shoot formation and combination of (BAP 1.0 mg/l+ Kin 0.5 mg/l) least response of shoot formation in Bizet (11 days) and Domingo (19 days). Shoot tip showed more pronounced effect for shoot formation than nodal cuttings. Welldeveloped shoots were shifted for their multiplication. Maximum number of multiple shoots were obtained on MS medium containing (1.0 mg/l BAP & Kin) and (BAP 1.0 mg/l + Kin 0.5 mg/l) in Domingo. These multiple shoots increased in their number when were given subsequent incubation period. Addition of Kinetin to BAP shows fair shoot multiplication response. Shoots after attaining the size of 5.0 to 7.0 cm were shifted for rooting. Best rooting response was obtained on Domingo (¼ MS Basal + NAA 1.5 mg/l), Bizet (½ MS Basal + IBA 1.0 mg/l) and combination of (NAA 1.0 mg/l+ IBA 1.0 mg/l) Baltico. Well rooted plants were shifted into glass house for hardening and acclimatization and were shifted to green house condition.
