Tomato leaf curl virus (ToLCV) disease is a serious threat for tomato cultivation in the tropics and subtropics. This disease can be managed by incorporating resistance genes in the cultivars. A Ty-2 CAPS marker developed by Peter Hanson and co-workers was validated in the parental lines. The marker was studied in the lines viz. DVRT-1, DVRT-2, H-24, H-86, Fla478, Punjab Chhuhara and in few backcrosses. After PCR amplification and restriction digestion the resistant amplicon (~200 bp) was detected in H-24, DVRT-1, DVRT-2 and surprisingly in Punjab Chhuhara as well. H-86 carried the susceptibility allele (~ 350 bp). This kind of marker is not diagnostic for Ty-2 gene and can be used for detecting Ty-2 gene in few crosses with known parentage.
