Forensic histology analysis of tetramethylene glycol effects based on histological changes

Abstract

Tetrametylene glycol is a precursor of gamma-hydroxybutyrate (GHB) act as GABAb agonist primarily affects the central nervous system and has depressant effects. This current experiment is conducted by the observation and examination of histological changes based on the toxicity effect of tetramethylene glycol in adult male Sprague Dawley rats. Sprague Dawley was administered with tetramethylene glycol by intraperitoneal route for consecutive 14 days. Total of rats (n=40) divided randomly into group of two (n=10), three (n=10), four (n=10) was treated with tetramethylene glycol 50 mg/kg/day (low dose), 300 mg/kg/day (high dose), and 500 mg/kg/day (extreme high dose) respectively. The histological examination was carried out by Haematoxylin & Eosin (H&E) staining. The toxicity effects of tetramethylene glycol display on histological changes shows the consequences of erythrocytes and hepatocytes under microscopic observation. In addition, these results showed direct proportionally relationship between concentration of tetramethylene glycol and histological changes on Sprague Dawley's tissue. In another words, the increases of tetramethylene glycol doses directly increasing the severity of histological changes. Principal Componet Analysis (PCA) was used to cluster the existence of cell after treatment with different dose of Tetramethylene glycol. It shows only, echinocyte and lymphocyte still available even after treated with 500 mg/kg Tetramethylene glycol.
