Evaluation of role of gapdh, GSK-3, B-catenin, DNA fragmentation and immunohisto chemistry of caspase 3 in estimation of post mortem interval in albino rats

Abstract

Background: Death leads to marked biochemical changes in tissues due to anoxia, degradation of cells with changes in enzymatic reactions and stoppage of anabolic processes in cells. Chemical markers help in accurate determination of post-mortem interval through determination of biochemical changes. Aim of Study: Our study was conducted for assessment of role of GAPDH, GSK-3, B-catenin, DNA fragmentation and immunohistochemistry of caspase 3 in estimation of post mortem intervals and evaluation of its accuracy. Method: In our study, Twelve Albino male rats were divided into 3 groups with 4 rats in each group. The scarified whole bodies were kept at 25+2°C room, for 0, 24 and 48 h. At the designed time of PMI, the organs samples taken after dissection are brain, heart and kidneys. They were frozen in liquid nitrogen for immunohistochemical examinations and biochemical study in kidneys, brains and hearts. Result and Conclusion: Biochemical study of Glyceraldehyde 3-phosphate dehydrogenase (GAPDH), glycogen synthase kinase-3 (GSK-3) and DNA fragmentation shows their importance in estimation of post mortem interval at time of death, after 24 h and after 48 h of death of Albino rats. But B-catenin has a limited role in estimation of post mortem interval in Albino rats. Immunohistochemically evaluation of caspase 3 in kidneys, brains and hearts of Albino rats at time of death has an important role in estimation of post mortem interval at the time of death, after 24 h and after 48 h of death of Albino rats. So, we recommend for further studies on their roles in human being estimation of post mortem interval. Keywords: GAPDH, GSK-3, B-catenin, DNA Fragmentation, Caspase 3, post mortem interval.
