In DNA fingerprinting the gender of a sample is determined by the presence of Y allele in amelogenin in autosomal STR. Failure in amelogenin-based gender typing of individuals have been observed worldwide. We used the AmpFlSTR Identifiler plus Kit and observed that out of 209 male profile ssuccessfully obtained, the profile of 208 samples was complete while only one profiles was observed without Y allele on amelogenin marker during the random routine case work analysis of the case exhibits deposited in Forensic Science Laboratory, Rohini, Delhi, India. The above one Y deleted sample was amplified by use of AmpFlSTR Y-Filer kit having 16 markers of Y chromosome. AmpFlSTR Y-Filer kit developed all other 16 markers of Y chromosomes for determiningthe male gender of sample.In this paper we review the role of AmpFlSTR Y-Filer Kit for the sex-determination in case of Y Allele deletion in forensic DNA samples.
