Development of analytical method on HPLC-UV for determination of lignocaine in forensic samples – blood, injection and viscera

Abstract

A highly, specific, scientific, accurate and rapid method in instrument High - Performance Liquid Chromatography (HPLC) was used to identify the presence of Lignocaine drug in the forensic samples – blood, injection and viscera (liver). The extraction of Lignocaineis done by liquid – liquid method. In the following study, a new method is developed in HPLC under modified chromatographic conditions of the mobile phase consisting disodium hydrogen phosphate: methanol: triethylamine in the ratio of 50:50:0.1 (v/v/v) where the flow rate has been set at 1.2ml/min. The range of wavelength was set at 220nm. The assay exhibited peculiar linearity range (r2>0.997) via graph interpretation of area v/s concentration whose value ranges from 10-40ppm. The HPLC-UV method is one of the most reliable and analytical method in order to determine and to form calibration curve from linearity as well as to determine accuracy, precision, % RSD, robustness and detection of LOD & LOQ.
