Human tissues preserved in fixatives by and large represent an essential component in medical diagnosis and therapeutic procedures employed to study the detailed progression of diseases, or are taken during autopsies for establishing the cause of death.These archived tissue specimens constitute an invaluable resource for histological examination, molecular diagnostic procedures and for DNA typing analysis in forensic investigations. However, the fixed available material is often limited in size and quantity. Moreover, the recovery of DNA is often severely compromised by the presence of covalent DNA– protein cross-links generated by formalin and its long term effects. In the present study we describe the evaluation of buffer formulations, sample lysis regimens and DNA recovery strategies incorporating different human tissue specimens preserved in formalin over variable periods of time for the extraction of DNA suitable for forensic investigations.
