Preparation of suitable papers for dna storage of blood samples for medical and forensic purposes compared with a specific standard prepared papers

Abstract

The goal of this study is to prepare papers suitable for storing DNA based on some chemicals, aimed for use in forensic laboratories, fieldwork, and scientific research with unexpansive and locally available materials. The Paperwhich is filter paper was treated with reagents prepared based on chemicals of different concentrations,then, 100 microliters of fresh blood are spotted on it. These papers were left to dry at room temperature before being stored to be extracted after different periods of time (one week, two weeks, one month, two months, and three months, four months, five months) using the Geneaid kit, then measuring the DNA concentrations using a Quantus fluorometer (Promega) and detecting the gene (IL37) using the polymerase chain reaction. And the results proved the efficiency of these manufactured papers to store DNA for five months. Statistical analysis was shown that reagents A and E have the ability to maintain DNA stability on paper at the best concentration in the fifth month. Also, the commercial FTA cards were used as a control and a paper filter that was not treated with solutions as a Blank.
