Isolation Purification and Analysis of Free Radical Scavenging Activity of Flavonids and Alkaloids from Indigofera longeracemosa Linn

Abstract

The Aim and Objective of the study is to isolate, purify and to analyze the free radical scavenging activity of Alkaloids andFlavanoids form Indigofera longeracemosa.

Fresh plants of I. longeracemosa were collected and authenticated in Botanical Survey of India of Tamilnadu Agricultural University. The samples were powdered and the extracts were prepared using petroleum ether, chloroform and methanol. Thereafter, the extracts were isolated and separated using TLC. Isolation of alkaloids and flavanoids was done using column chromatography. Purification of the compounds was done using preparative TLC and the analysis of phenolics and antioxidant activity were measured. The methanolic extract was profiled using HPLC. All the separated compounds were screened for the antioxidant activity.

The phenolic content of the dried plant material was 63.5% mg equivalent of gallic acid which reveals that the drug which we investigated was a flavonoidal drug. In the antioxidant activity of isolated compounds, all the compounds were found to possess antioxidant activity. All the readings were above the control reading which confirms the activity of the compounds. In the HPLC profiling of the methanolic extract, among the major compounds the compound with Rt 6.25 was the major one which comprising around 18.35% of the total constituents.

These inferences tell that, the plant extract of I. longeracemosa shows satisfactory results of antioxidant activity. There should be further research to study the activity of I. longeracemosa in depth.
